The hsa-mir-200c_3DDSD_s (BBa_K5086114) is formed by the combination of hsa-mir-200c_3DDSD (BBa_K5086024), hsa-mir-200c_target (BBa_K5086001), and three wt (BBa_K5086017) chains. Among them, the sequence at the 5' end of hsa-mir-200c_3DDSD is successively linked to the hsa-mir-200c_target and the three wt chains in a complementary pair, and the sequence at its 3' end is complementary to the chain of Universalness_h (BBa_K5086009), thus hsa-mir-200c_3DDSD_s can bind specifically to the Universalness_h strand thus accomplishing the recognition of the input signal. DNA polymerase polymerizes DNA in the 5'-3' direction starting from the 3' end of the target hsa-mir-200c_Rep. During DNA polymerization by Bst DNA polymerase, the wt strand is displaced and released by the newly synthesized daughter strand, and a single strand of DNA with sequence identical to that of the target strand is also released. During the DNA polymerization by Bst DNA polymerase, the wt strand will be replaced and released by the newly synthesized daughter strand, and also release the DNA single strand with the same sequence as that of the target strand (U to T), which will act as a target to undergo the first step of the polymerase-mediated strand replacement reaction again to realize the self-enhancement of the DDSD process, and the wt strand will be involved in the subsequent reaction to produce the fluorescence report.
